The Sherlock Holmes star will take on the role of a recovering drug addict called Nicole Devlin who befriends Denzel Washington's pilot Whip, Deadline reveals.
Flight follows an investigation into an emergency landing by Whip, who is hailed a hero for saving the lives of 98 out of 106 passengers. However, an investigation into the crash reveals that Whip's behaviour the night before may be to blame for the incident.
Gnome Attack director Zemeckis will be directing from a script by John Gatins. Flight will be Zemeckis's first live-action feature since the 2000 survival drama Castaway.
Reilly, who has also appeared in Eden Lake, is in contention with Olivia Wilde and Moon star Dominique McElligott for the part.
Reilly will be seen reprising her role as Mary Morstan in Guy Ritchie's follow-up Sherlock Holmes: A Game of Shadows.
In July, Ritchie said that it had been an easy decision to make the sequel to the hit adaptation, starring Robert Downey Jr and Jude Law.
