The actress will play Billy Crystal's estranged daughter, who lands him and her mother with her three grandchildren, according to Variety.
Us and Them deals with the generation gap and the couple's reconciliation with Tomei's character. Crystal will be appearing opposite Bette Midler, who takes on the role of his wife.
You Again's Andy Fickman is directing from a script by Tooth Fairy and City Slickers' writing team Lowell Ganz and Babaloo Mandel.
Tomei will next be seen in George Clooney's fourth directorial effort The Ides of March.
She will appear alongside Ryan Gosling, Evan Rachel Wood, Paul Giamatti and Philip Seymour Hoffman in the political drama, which sees Clooney star as a governor keen to get into the White House.
Tomei is also taking part in a one-night only reading of Dustin Lance Black's 8 with Morgan Freeman and Stand By Me director Rob Reiner.
