Stephen Soderbergh will direct the reboot of the original 1960s TV series, with original star George Clooney forced to quit production due to a back injury.
True Blood star Alexander Skarsgård is the other name on Warner Bros' list of potential lead actors, according to The Wrap.
The studio is set to be seeking "a leading man in his 20s" for the main role of agent Ilya Kuryakin, originally played by David McCallum.
The original TV series centred around U.N.C.L.E. (United Network Command For Law Enforcement), a secret international intelligence agency.
Soderbergh's latest film Contagion is released in the UK on October 21.
