The Dances with Wolves star was originally linked to the film back in July, but "scheduling conflicts" have caused him to leave the project.
"And in another interesting piece of casting news, Kevin Costner has had to pass on Django Unchained due to scheduling conflicts," Variety reporter Justin Kroll tweeted earlier this week.
The film stars Jamie Foxx in the titular role as a former slave who sets out to rescue his wife from a plantation owner, portrayed by Leonardo DiCaprio.
Inglourious Basterds star Christoph Waltz and Pulp Fiction star Samuel L. Jackson are also part of the film's cast.
Earlier this year, Costner was cast as Pa Kent in Zack Snyder's superhero reboot Superman: Man of Steel opposite Henry Cavill, Diane Lane and Michael Shannon
Django Unchained is currently scheduled to reach cinemas in December 2012.
