The former Prison Break and Heroes actor will play a spirit known as a 'deado' in the film, alongside Ryan Reynolds and Jeff Bridges, according to The Hollywood Reporter.
The film centres around a murdered cop (Reynolds) who is recruited for the Rest In Peace Department, a police force of ghosts who fight against evil spirits.
Kevin Bacon, James Hong, Marisa Miller, Mary-Louise Parker and Stephanie Szostak will also appear in the film.
R.I.P.D. will be directed by RED's Robert Schwentke.
Knepper recently starred in NBC's pilot episode of Reconstruction, a series about a soldier who travels across the country after the Civil War.
