The 66-year-old actress blamed producers for not creating any roles for women in the adaptation of John Le Carré's 1974 spy novel.
Gimme Gimme Gimme's Kathy Burke is the only female star in the movie, which also features Colin Firth and Benedict Cumberbatch.
Speaking to The Sunday Times, Mirren declared: "How many women were in that? I mean, come on. There weren't any women in the '70s?
"The secret service always had a huge number of women working for them, and they played major roles in real life. But they were ignored for this film!"
Tinker, Tailor, Soldier Spy, directed by Tomas Alfredson, topped the UK box office last week.
