The actress - who has recently appeared in The Help, The Debt and The Tree of Life - will play Cruise's love interest in the film, according to Variety.
It was reported last month that Chastain was one of five actresses in contention for the part, alongside The Girl with the Dragon Tattoo's Noomi Rapace, Olga Kurylenko (Quantum of Solace), Brit Marling (Another Earth) and Cowboys & Aliens' Olivia Wilde.
Previously given the working titles of Horizons and Oblivion, the Universal Studios project is being directed by Tron: Legacy's Joseph Kosinski with a script adapted from his own graphic novel Oblivion.
Cruise's character in the film has responsibility for preventing an alien attack on a post-apocalyptic Earth.
Production on the project is due to start in the winter ahead of a projected release date of July 2013.
