Tim Daly and Kevin Conroy - the long-time voices of Superman and Batman - will also return for the new feature, reports TV Guide.
Michael Rosenbaum, Susan Eisenberg and Carl Lumbly will also reprise their roles as The Flash, Wonder Woman and Martian Manhunter.
Justice League: Doom, which was announced at WonderCon, is based on Mark Waid, Howard Porter and Drew Geraci's 2000 JLA story arc 'Tower of Babel'.
The story focuses on an apparent betrayal by Batman, whose secret files on the weaknesses of the Justice League fall into their enemies' hands.
Conroy, Rosenbaum, Eisenberg and Lumbly all featured in the Justice League Unlimited animated series. Rosenbaum also played Lex Luthor in Smallville.
Justice League: Doom was executive produced by Bruce Timm and directed by Lauren Montgomery.
It is the final piece of work by writer and producer Dwayne McDuffie, who died on February 22.
The Justice League: Doom trailer will debut at New York Comic Con on October 14.
