The Terms of Endearment actress will accept the prize at a special ceremony to be held in LA on June 7, 2012. TV Land will air the ceremony later in the month.
Howard Stringer, head of AFI's board of trustees, praised MacLaine as one of the silver screen's most iconic stars.
Stringer said: "Shirley MacLaine is a powerhouse of personality that has illuminated screens large and small across six decades. From ingénue to screen legend, Shirley has entertained a global audience through song, dance, laughter and tears, and her career as writer, director and producer is even further evidence of her passion for the art form and her seemingly boundless talents."
The actress follows in the footsteps of Morgan Freeman, who was this year's recipient of the prestigious AFI honour.
MacLaine won an Academy Award for her role in Terms of Endearment and also earned nominations for Some Came Running, The Apartment and The Turning Point, among other films.
She was given the French Legion of Honour by the country's Culture Minister Frederic Mitterrand in September.
