The Dark Knight actor will play the Beach Boys legend in Randy Miller's The Drummer.
Eckhart had been in negotiations to play the musician for several months, and will executive produce the film. He will also record cover versions of tracks from Wilson's 1977 album Pacific Ocean Blue.
"We'd been trying to figure out who the right person would be to fill this role," Miller told Variety. "It's quite a demanding role so it had to be a great actor who could sing."
The movie will document the last six years of Wilson's life. The drummer died in 1983 after years of alcohol abuse.
"There's a discovery about Dennis with this film," Miller added. "Everybody knows Brian [Wilson], but nobody really knows Dennis's music."
The Beach Boys recently unveiled details of their upcoming SMiLE Sessions box set.
