According to Collider, the Friends with Benefits star is set to accept a co-lead role in the film, which is about the '60s folk music scene in New York City.
Earlier this month it was reported that Carey Mulligan was close to signing up for a role opposite her Drive co-star Oscar Isaac.
Isaac is on board to portray troubled musician Llewyn Davis, while Timberlake would play a musician married to Mulligan's character.
No Country for Old Men producer Scott Rudin will reunite with the Coen Brothers for the film.
Inside Llewyn Davis is expected to be released sometime in 2012.
