The Debt actress is in line to play Hitchcock's wife Alma Reville, according to The Hollywood Reporter.
The adaptation of Stephen Rebello's non-fiction tale of the making of the classic movie has been in the works for several years.
It was initially being developed at Paramount by Glee co-creator Ryan Murphy, who acquired Anthony Hopkins for the lead role.
Hopkins remains attached, but the film is now being helmed by Anvil! The Story Of Anvil director Sacha Gervasi for Fox Searchlight.
The script has been adapted by Rebello and Black Swan co-writer John McLaughlin.
Sources have said that Mirren is still in the early stages of negotiations for the role.
Hitchcock struggled to finance Psycho independently after no studio would get involved with the 'lowly' horror movie.
Alfred Hitchcock and the Making of Psycho is rumoured to begin shooting in April.
