The 41-year-old actress is being awarded for her role as Minny in The Help with the festival's 'Breakthrough Performance' prize.
The Palm Springs International Film Festival chairman Harold Matzner commented that Spencer is receiving the prize because she helped bring life to one of the most critically acclaimed films of the year.
Matzner said in a statement: "Octavia Spencer is a consummate actress who seamlessly blends her dramatic and comedic talents, creating performances of uncommon depth.
"As the irascible Minny in The Help, she captures the essence of a woman who deals with life head on, relishing confrontation and serving as a source of courage and humour for those whom she loves."
Freida Pinto, Jennifer Hudson and Marion Cotillard are among the previous recipients of the 'Breakthrough Performance Award'.
The Palm Springs International Film Festival will also give a career achievement award to Glenn Close and the 'Desert Palm Achievement Actress' trophy to Michelle Williams when it kicks off in January.
