The 50/50 actor starred in the 2008 stoner comedy opposite James Franco, and has hinted that a new outing may be produced in the future.
"I don't think it's so precious that I wouldn't want to ruin it with a sequel," he told E! Online.
"So yeah, I would be [interested]. We talk about it. We joke around about it."
Rogen's writing partner Evan Goldberg revealed earlier this year that Pineapple Express 2 may be in the works.
He explained at the time that they were thinking of making a crossover with Superbad character Fogell, otherwise known as 'McLovin', played by Christopher Mintz-Plasse.
He said: "Well, we toyed with that idea right after Pineapple came out. We thought that people would enjoy McLovin and Franco together; but we're never going to touch Superbad ever again, that's a terrible idea."
Seth Rogen will host next year's Independent Spirit Awards on February 25, 2012.
