The actress will join Jude Law, Blake Lively and Channing Tatum on the medical drama, reports Deadline.
Zeta-Jones and Soderbergh previously collaborated on 2000's Traffic, for which she won a Golden Globe.
Screenwriter Scott Z Burns (Soderbergh's collaborator on The Informant!, Contagion) is writing the film, which deals with psychopharmacology, the study of the effect of drugs on the human mind.
Zeta-Jones and Law will play doctors, with Lively as a woman addicted to prescription drugs and Tatum her husband, who is due to be released from prison.
Tatum collaborated with Soderbergh on Haywire and the forthcoming Magic Mike, and Law appeared in Contagion. Lively is the only star not to have worked with the director in the past.
