The film, titled The Partner and based on the best-selling book of the same name, will be a thriller about a lawyer who fakes his own death, according to Deadline.
Several Grisham novels have been turned into successful movies, including the 2003 John Cusack vehicle Runaway Jury.
Hancock's last film, 2009 drama The Blind Side, earned a 'Best Picture' nomination and a 'Best Actress' Oscar for Sandra Bullock at the Academy Awards.
Hancock will also pen the screenplay for the adaptation, but there is no word as yet on the cast of the film.
