The Oscar-winning actor, who has been linked to the project since last year, joins Harrison Ford and Asa Butterfield in Summit Entertainment's adaptation of Orson Scott Card's novel.
Ender's Game, written and directed by Gavin Hood, tells the story of a child prodigy who is drafted into the military to help save the human race from an alien threat. Kingsley will portray a war hero named Mazer Rackham in the film, Reuters reports.
Mr Young star Brendan Meyer joined the cast back in December 2011. Brandon Soo Hoo, Hailee Steinfeld and Abigail Breslin will feature in supporting roles.
Card has written numerous follow-up books set in the Ender's Game universe, several of which have been adapted as graphic novels by Marvel Comics.
Ender's Game enters production next month in New Orleans.
