Anton Corbijn will direct the spy thriller, which is based on Andrew Bovell's screenplay adapted from the John le Carré novel of the same name.
A Most Wanted Man takes place in modern-day Hamburg, Germany and features themes of the international war on terror and money laundering. The story follows an enigmatic Chechen ex-prisoner with a claim to a fortune held in a private bank.
Ides of March star Hoffman is in line to play a roguish chief of a covert German spy unit named Bachmann, who is keeping a close watch on the Chechen.
"We are completely thrilled at the prospect of Philip Seymour Hoffman playing Bachmann in Andrew Bovell's brilliant adaptation of this very modern thriller by John le Carré," said producer Gail Egan.
"We will start shooting in Hamburg in September and are delighted to be working with Anton Corbijn on this film. His unique vision for le Carré's complex and stylish story is very contemporary, very real, very human."
A Most Wanted Man is expected to arrive in cinemas in 2013.
