From Up on Poppy Hill was the highest-grossing Japanese film of 2011, earning over $56 million (£35m).
The English version is set for a theatrical release in the US and the UK next year.
Collider reports that the legendary animation studio has signed up a star-studded voice cast for the new dub, including Gillian Anderson, Jamie Lee Curtis, Christina Hendricks, Ron Howard, Emily Osment and Aubrey Plaza.
Set in Yokohama in 1963, the film centers on Umi (Sarah Bolger) and Shun (Anton Yelchin) and the romance that develops between them as they join forces to save their high school's clubhouse from demolition.
The film is written by Ghibli founder and Spirited Away director Hayao Miyazaki, and directed by son Goro.
