Movie producer Scott Rudin, who recently worked on The Queen, has paid $2 million (£990,000) for the rights to Robert Graves' 1934 novel.
Rudin is said to have asked The Departed screenwriter William Monahan to pen the script.
The BBC adapted the novel, which tells the story of the Roman Empire through the eyes of Claudius, a member of the imperial family who suffers from a stutter and a limp, into an Emmy-winning mini-series in 1976.
Sir Derek Jacobi won a BAFTA for his performance in the title role.
