According to Reuters, Pegg will play Enterprise engineer Scotty, while Cho will appear as Sulu, the character who was made famous by George Takei.
It was revealed on Wednesday that Hulk actor Eric Bana will play the villain Nero in the 11th movie in the franchise.
Directed by JJ Abrams, the movie is expected to take place in the early years of the Enterprise's crew.
The movie will reportedly begin shooting in November and is expected to be released for Christmas 2008.
