Day-Lewis scooped the Best Actor gong for his role as a Texan oil prospector in There Will Be Blood.
He dedicated the award to the late Heath Ledger, saying: "In Brokeback Mountain, he was unique, he was perfect. That scene in the trailer at the end of the film is as moving as anything I think I've ever seen."
Julie Christie won Best Actress for her portrayal of an Alzheimer's sufferer in Away From Her, while American Gangster star Ruby Dee beat favourite Cate Blanchett in the Best Supporting Actress category.
Javier Bardem picked up the Best Supporting Actor prize for No Country for Old Men, which also won Best Picture and Best Ensemble Cast.
The ceremony was the first big awards bash of the season after the Golden Globes show was replaced with a press conference due to the ongoing WGA strike.
The SAG was granted a waiver in return for the guild's support of the writers' action.
