The World War II drama beat The Lives of Others, No Country for Old Men, American Gangster and There Will Be Blood to claim the top prize at the London ceremony, hosted by Jonathan Ross.
Daniel Day-Lewis scooped the Best Actor gong for his turn as a Texan oil prospector in There Will Be Blood, while Marion Cotillard beat Julie Christie to win Best Supporting Actress for her portrayal of singer Edith Piaf in La Vie en Rose.
Javier Bardem was named Best Supporting Actor for No Country for Old Men, for which Joel and Ethan Coen also won Best Director.
Michael Clayton's Tilda Swinton triumphed in the Best Supporting Actress category at the event, which also saw Sir Anthony Hopkins receive an Academy fellowship.
Famous names handing out trophies included Sylvester Stallone, Cuba Gooding Jr, Eddie Izzard, Jeff Goldblum, Kevin Spacey and Sir Ian McKellen.
