The film will reunite the pair with director Nick Stoller and star Jason Segal, who worked together on the upcoming Forgetting Sarah Marshall.
The story focuses on a man (Segal) and his fiancée as they navigate the ups and downs of their five-year relationship.
"It's definitely an extension of our desire to explore the depth of human misery," revealed Stoller. "If Sarah Marshall (Heroes' Kristen Bell) and Jason's character had stayed together, this might be the sequel."
Stoller and Segal will co-write the script. The pair met while working on Apatow's TV show Undeclared.
