The actor had signed to play a gay cheerleading instructor in the movie, but pulled out "three weeks to a month ago", reports OK! USA.
Doctors confirmed last week that he has been diagnosed with pancreatic cancer.
His spokesperson Annett Wolf previously insisted that Swayze was "continuing his normal schedule", but it has now emerged that he will not return to work in the near future.
He has reportedly been replaced in the movie by The Break-Up actor John Michael Higgins.
Swayze is currently recovering at home after undergoing a third round of chemotherapy.
