The Shrek star will front the Los Angeles ceremony on June 1, which includes gongs for Best Screen Kiss and Best Villain.
Talking about the pressures of the job, Myers said: "Hosting the MTV Movie Awards is like a party, but without having to do beer runs in your mum's mini van.
"We do beer runs in Will Smith's four-storey motor home."
Myers hosted the awards once before in 1997, when winners included Tom Cruise and Claire Danes.
Will Smith, Lindsay Lohan, Sarah Jessica Parker and Jessica Alba have hosted the ceremony in the past.
Last year's presenter Sarah Silverman caused controversy by mocking Britney Spears and her children.
