The Sweeney Todd director will reportedly helm the Warner Bros film, which is set up at Johnny Depp's Infinitum-Nihil production company. John August will write the movie.
Dan Curtis's Dark Shadows ran on US TV from 1966 to 1971. The series's popularity surged when vampire Barnabas Collins was introduced a year into its run.
In an interview with Entertainment Weekly last year, Depp stated: "When I was a kid I wanted to be Barnabas.
"He was super-mysterious, with that really weird hairdo and the wolf's-head cane."
Burton, August and Depp have previously collaborated on Corpse Bride, Charlie And The Chocolate Factory and Big Fish.
