The boyband will perform their new single 'Greatest Day' at the Liverpool Echo Arena on November 6.
Gary Barlow, Mark Owen, Jason Orange and Howard Donald last played the EMAs at the inaugural ceremony in 1994, when they won the Best Group gong.
"This is a historic moment in the history of the MTV Europe Music Awards," said the event's executive producer Richard Godfrey.
"Take That were part of the first ever Awards show and it is only fitting that they return to the MTV Europe Music Awards for its fifteenth anniversary."
Other performers confirmed include Beyoncé, Pink, Duffy, The Killers and Kid Rock. American singer Katy Perry will helm the show.
Take That confirmed last week that their new album The Circus would be released on December 1.
