The singer has agreed to appear at the top London venue as part of a whirlwind 24 hour visit to Britain.
"It has taken months of very patient planning but the hard work has been more than worth it," a source told The Sun.
Previous acts to perform there include Kylie Minogue, Mariah Carey and Christina Aguilera.
Spears will perform on this week's X Factor before releasing her new album Circus on December 2.
