The Las Vegas band will play the final night of the four-day festival on July 19.
"I'm very pleased to have the Killers headline this year. Not only is it their first time with us, but they are joining an amazing bill for our 15th anniversary. This year's festival is shaping up to be the best in Europe," founder Vince Power said.
Oasis recently announced that they would headline on the opening night on July 16, with Kings of Leon and Franz Ferdinand playing the main stage on the second and third evenings respectively.
Last year's festival saw over 50,000 revellers flock to the Costa del Azahar each day. Previous performers include Radiohead, Depeche Mode, Brian Wilson, Blur, The Strokes and Lou Reed.
Benicassim festival takes place between July 16 and July 19.
