The band's slot on the Saturday night is now listed on the tour plans posted on their website and MySpace.
Alex Kapranos's group first played the festival in 2004 on the Other Stage. They returned last year to perform a last-minute gig on The Park Stage.
Other acts expected to play the main stages this year include Neil Young, Bruce Springsteen, Blur, The Ting Tings, Lily Allen, Kaiser Chiefs, Rolf Harris, Kanye West and BB King.
Glastonbury organisers confirmed earlier this week that all tickets had sold out for the event.
