Their set at the event, which will be the Stereophonics's only UK festival appearance in 2009, is expected to feature hits from their recent 'Best Of' compilation and new material.
"The Isle of Wight festival is gonna be a great weekend," said frontman Kelly Jones.
"We're recording most of the first half of this year so to step out of the studio and headline the festival this year with it being even bigger than when we played it a several years ago, will be a highlight for us! It's gonna be rocking. Enough to make a bulldog wanna snap his chain!"
Co-headliners Razorlight will be playing the event for the first time since 2002.
The Friday lineup was confirmed in January and will feature The Prodigy, Pendulum, Basement Jaxx and The Ting Tings. It was also revealed earlier this month that Manchester baggy veterans The Charlatans will be performing and curating the Big Top at the three-day event.
Isle Of Wight takes place on June 12, 13 and 14 at Seaclose Park, Newport. Tickets are currently on sale.
