The Grammy winners' hit single overtook Leona Lewis's 'Bleeding Love' by passing 547,000 online sales, says The Sun.
The Nashville band also hold the title for the biggest-selling digital album ever in the UK with Only By The Night.
Columbia managing director Mike Smith said the band had "dominated" rock music in the UK over the past year. "In any format Only By The Night would be acknowledged as one of the most important albums of the decade, but it has truly come into its own in the digital age," he said.
"Despite only being released five months ago it is now the biggest-selling digital album of all time in the UK. In addition, this week 'Sex On Fire' became the single biggest downloaded track since the birth of the UK digital chart."
