Black Francis's band will play the main stage on the same day as Neil Young, who closes the three-day event.
Formed in Boston in 1986, the four-piece achieved critical acclaim for albums such as Surfer Rosa, Doolittle and Bossanova. They split in 1993, but reformed five years ago and played shows at Coachella and Reading/Leeds festivals.
"It was hard to think of someone capable of complementing the musical talent of Neil Young but once I had the idea it was blindly obvious and it will surely be a legendary day on the Isle of Wight," said festival promoter John Giddings.
The Isle of Wight festival takes place at Seaclose Park, Newport on June 12, 13 and 14. The Prodigy, The Charlatans, The Ting Tings, Stereophonics, Razorlight and Basement Jaxx are all confirmed to perform.
