Tickets for the show, which takes place on Friday, June 26, go on sale this Friday at 9am.
The Kooks will be the main support act, with more bands still to be announced. Bruce Springsteen and Neil Young are headlining the other two days at the event.
"Going to three days was a big step for us and Hard Rock, and then the Killers came along and in my book, that's a no brainer," said Live Nation's John Probyn.
"We are committed to bringing great music to the heart of London every summer and with such an amazing line up this year, there is something for everyone."
