The two acts will open the concert at Manchester Evening News Arena on June 26, a week before Blur play two gigs in London's Hyde Park.
"Every show Blur play this summer is going to be unique and different, so we're delighted Klaxons and Florence and the Machine have agreed to join us for the evening," Blur manager Chris Morrison said.
"Blur are really looking forward to this gig – they've always had an amazing reception in Manchester."
Speaking to NME, Britis Critics' Choice award winner Florence Welch added: "I'm really excited about supporting Blur in Manchester. I can't comprehend that it is happening, to be honest!"
