The rapper performed a 30-minute set to more than 2,000 fans at the Motorcity Casino's Soundboard stage last night.
Speaking to the packed audience, he admitted that he was a "little bit nervous because I don't have my right-hand man", a reference to the late MC Proof.
He also joked about his well-documented battles with drink and drugs, saying: "This might actually be one of the best shows I've ever done, because when I get offstage I'm actually going to remember it!"
His set consisted of seven songs taken from his current Relapse LP, including singles 'We Made You' and 'Crack A Bottle', and an encore of his 2002 track 'Lose Yourself', featuring his D12 bandmates.
Eminem had earlier used his Twitter account to give away the last remaining tickets for the show, directing fans to specific people or places in the city, including a mailbox, a restaurant and the inside of a shoe.
The entire concert was filmed and two of the tracks will be shown on Jimmy Kimmel Live this Friday.
