The rapper took the top spot on the publication's annual 'Power 30' list.
The '99 Problems' star's clothing label Rocawear, co-ownership of the New Jersey Nets and Live Nation deal all contributed toward his hip-hop heavyweight status.
Sean 'Diddy' Combs, 50 Cent, Lil Wayne and Snoop Dogg also made the top thirty, while US President Barack Obama was awarded with an honorary mention for his influence on aspiring rappers.
Earlier this week, it was reported that Jay-Z's new album has a rumoured release date of September 11.
