The indie-dance crossover group will fill the special guest slot on the Park Stage at 7.50pm on June 27, NME reports.
The announcement follows news that N.E.R.D. had been added to the Pyramid Stage lineup for June 26.
Klaxons recently revealed that their uncompleted second album would be delayed for a 2010 release.
The Glastonbury Festival takes place this weekend in Pilton, Somerset and is headlined by Neil Young, Bruce Springsteen and Blur.
