Frontman Bono dedicated their hit 'Angel of Harlem', which was written about Billie Holiday, to the 'Thriller' singer. He also added lines from Jackson's hits 'Man In The Mirror' and 'Don't Stop 'Til You Get Enough' at the end of the performance.
Before the track, the Irish singer told the Spanish crowd: "We met Michael Jackson many times over the years and he was an unspeakable talent."
Later in the show, Bono put on a Barcelona football shirt, applauding the team for using charity UNICEF as a sponsor.
U2 kick off the UK leg of their latest tour at Wembley on August 14. They will also play Cardiff, Glasgow and Sheffield.
The setlist in Barcelona included hits 'Beautiful Day', 'The Unforgettable Fire', 'Sunday Bloody Sunday', 'With Or Without You' and 'One'.
