Their hit 'I Gotta Feeling' - currently top of the US, Australian and Irish charts as well - climbs from three to one this week.
It matches the success of the group's previous single 'Boom Boom Pow', which spent two non-consecutive weeks at number one in May and June.
Behind Black Eyed Peas, JLS slip to two with 'Beat Again', while Mr Hudson and Kanye West fall to three with 'Supernova'.
Meanwhile, US rapper Pitbull jumps from seven to four with 'I Know You Want Me (Calle Ocho)'.
The highest new entry of the week comes from Torchwood actor John Barrowman, who debuts at 14 with his version of Barry Manilow's 'I Made It Through The Rain'.
