Fans delayed the start of Saturday night's concert by singing in both English and Polish, as well as performing traditional birthday song 'May You Live A Hundred Years'.
The 51-year-old's voice wavered as she told the crowd: "Every time I do a show, and every time I look into your eyes and every time I see your smiles, you changed my life.
"It's amazing that I've been able to do what I do for so many years, thank God, I love my job, I love being able to be here with you tonight."
She added: "This is the best birthday present ever, so thank you."
Madonna was previously criticised by religious groups for scheduling the Polish date on a Roman Catholic holiday celebrating the heavenly assumption of the Virgin Mary.
According to the BBC, a small group of people held up protest banners outside the concert venue.
