'3', a new track taken from her upcoming Singles Collection, sold 255,000 downloads in its first week of release to debut at the summit of the Billboard Hot 100.
The singer previously reached the top with 'Womanizer' last year and '...Baby One More Time' in 1998.
Behind Spears, Jay Sean falls to two with 'Down', while Miley Cyrus slips to three with 'Party In The USA'.
Notable gains outside the top ten come from Black Eyed Peas, who jump from 32 to 17 with 'Meet Me Halfway', and David Guetta, who climbs from 34 to 25 with Akon collaboration 'Sexy Chick'.
