The Phoenix Nights comic's latest project will premiere on BBC One on November 20. The video will include 100 British and American celebrities, according to The Sun.
"It's taken two years of planning to create the video and I just know people are going to be both astonished and delighted," said Kay. "It's been a real labour of love but the result is joyous, potentially another 'Amarillo'!"
Kay has previously released covers of '(Is This The Way To) Amarillo' and 'I'm Gonna Be (500 Miles)' for Comic Relief, both of which reached number one. He also scored a hit with his comedy alter ego Geraldine McQueen last October, landing the top spot with spoof single 'The Winner's Song'.
