Rock journalist David Fricke will host the rare event, part of the 'LIVE from the NYPL' series, reports Reuters.
The reunion, taking place on December 8, was spurred by the recent publication of The Velvet Underground: New York Art, which collects previously unseen photographs, artwork, Reed's handwritten music and lyric notes, plus press clippings and other memorabilia.
The occasion will mark the bandmates' first public appearance together since the group's induction into the Rock and Roll Hall of Fame in 1996.
The original Velvet Underground lineup included Reed, Tucker, John Cale, Sterling Morrison and sometimes vocalist Nico.
The affair will happen at the Celeste Bartos Forum in the NYPL's Stephen A. Schwarzman Building. Tickets reportedly sold out minutes after the event was announced.
