The 'Where Did All The Love Go?' rockers are the first of three headliners to be revealed for the Scottish event, which takes place near Kinross from July 10 to July 12 next summer.
"T In The Park holds a special place in our hearts," said frontman Tom Meighan. "We've always said how much we'd love to headline it and now we are. We can't wait, we're going to play a blinder and it's going to be massive for us and for the crowd."
Geoff Ellis from DF Concerts added that Kasabian's live reputation helped secure them the headline slot. "I'm chuffed to bits that we can announce one of our headliners this early on," he said. "Kasabian blow me away every time I see them and they're going to be amazing headliners. We've had lots of fans tell us they want to see Kasabian headline T In The Park so it's great to be able to give them an early Christmas present!"
A limited number of earlybird tickets for the 2010 event go on sale tomorrow at 9am.
