All four bands will take part in the travelling Sonisphere Festival, starting on June 16 at Bemowo Airport in Warsaw, Poland, reports Billboard.
The groups are currently confirmed to play together at single dates in Warsaw and Prague, with talks ongoing for other appearances.
The Sonisphere Festival also includes two-night stands in Sofia, Bulgaria, Bucharest, Romania, Istanbul, Turkey and Knebworth, England.
"Who would have thought that more than 25 years after its inception, thrash metal's big four would not only still be around and more popular than ever, but will now play together for the first time?" said Metallica drummer Lars Ulrich in a statement.
Megadeth frontman Dave Mustaine called the forthcoming jaunt "a once-in-a-lifetime opportunity for heavy metal 'fans' to see the four greatest heavy metal 'bands' in American history back together on one stage".
Slayer and Megadeth will first team up for the 26-date American Carnage tour of North America, kicking off in January 2010.
