Fox
Journey cover 'Don't Stop Believin', which was featured in the series trailer and final scene, is on course to debut inside the Top 20.
The original version, currently at number seven in the countdown, has climbed inside the Top 5, according to midweek sales figures.
Meanwhile, renditions of Amy Winehouse's 'Rehab', Kanye West's 'Gold Digger' and 'Take A Bow' by Rihanna are expected to make an appearance in the chart.
"There is no doubting the impact that Glee is going to have on the chart this week, and possibly over the coming weeks," Official Charts Company managing director Martin Talbot said in a statement.
"Glee is a new cultural phenomenon - and the music charts are reflecting this, just as they did in the US last year."
Glee attracted an audience of 1.3 million when it premiered on E4 on Monday.
