The Welsh group will play the 32,000-seat venue, which was opened last July, on June 5.
"We want to make it a day to make Welsh people proud," frontman Kelly Jones said. "We will be the first band to play the stadium and we love breaking in venues, even dating back to 1993, being the first band to play the Ivy Bush in Cwmaman!"
Stereophonics previously played a record-breaking 13 sell-out shows at the Cardiff International Arena and most recently performed at Cardiff Castle in September.
Tickets for the Cardiff City Stadium gig go on sale on January 22 and are priced at £35.
