The Scottish singer, who found fame through Britain's Got Talent, was described as a unique talent by the hip-hop heavyweights.
According to The Sun, West said: "There is no doubt Cheryl Cole is hot but there is one hotter - Susan.
"There are loads of talented, good-looking girls who can sing and dance in the charts but there is only one Susan Boyle. I have never seen anything like her."
50 Cent added that he "probably would" collaborate with the 48-year-old. "We got really tough competition - now that the competition is Susan Boyle," he said.
Ealier this month, Prince William revealed that he has Boyle's debut LP I Dreamed A Dream on his iPod. Other famous fans of the singer include Lady Gaga, Demi Moore, Oprah Winfrey and Barack Obama.
